APP/PS1 mice
APP/PS1 double transgenic mice are transgenic animals generated using gene targeting technology. This animal can express mutated human amyloid precursor protein (APPswe) and human presenilin 1 (DeltaE9) fusion protein, and the expression of both genes is initiated by the mice prion protein promoter. The DeltaE9 mutation of the human presenilin gene is caused by the deletion of the ninth exon of the gene, which leads to early-onset Alzheimer's disease.
Features:
At 4 months of age, senile plaques are visible in the brain, and at 6 months of age, ΑΒ deposits are found in the cerebral cortex.
Compared with wild-type counterparts, memory-related behavioral defects appear at 10-12 months of age.
Applications
Study of human and mouse homologues of genes related to Alzheimer's disease.
Neurobiological research on Alzheimer's disease and neurodegenerative changes.
